Unlock Single Battle mode characters:
If you have a saved game file from Bleach: Heat The Soul 4 on your Memory
Stick Duo, just go to the Appendix menu and choose the "Convert" option.
Then, go to character selection screen in Single Battle mode and press
L or R to select any of the new characters.

Unlocked by beating him and Hiyori in Tag Master mode.

Senna:
Unlocked by beating Tag Master #16.

Seele Schneider Ishida:
Unlocked by beating Tag Master #8.

Unlocked by beating Ikkaku/Kempachi in Tag Master mode.

Unlocked by beating him and Rukia in Tag Master mode.

Unlocked by beating Ichigo and Hollow Ichigo in Tag Master mode.

Unlocked by beating Tag Master #14.
